Development of a modified hard gelatin capsule for colon-targeted drug delivery of hydrogel-based piroxicam microparticles

Purpose: To develop modified hard gelatin capsules (MHGCs) for colon-specific delivery of hydrogel-based piroxicam microparticles.
 Methods: Solvent evaporation technique was adopted the microencapsulation using liquid paraffin (PL.MPs) and soybean oil (PS.MPs) which were subsequently encapsulated in MHGCs (water-impervious). Anti-inflammatory vitro dissolution studies conducted on unencapsulated microparticles (MPs). Furthermore, sequential drug release from impervious carried out 2 h at pH 1.2 3 6.8, ≥ 5 7.4, to simulate stomach, small intestine, colon environment, respectively. Differential scanning calorimetry (DSC) analysis also formulations.
 Results: Edema inhibition PL.MPs PS.MPs within range 51.0 – 64.0 58.0 69.0 %, In revealed absence, minimal, highest amounts 1.2, 6.8 7.4 media, Decreased crystallinity indicated by a broad endothermic peak.
 Conclusion: The hold promise as potential alternative system designed colon-targeted delivery. However, clinical development are required.
